Site-directed mutagenesis in vitro by isothermal amplification

Аннотация. Методы изотермической амплификации ДНК получили широкое распространение в клинической диагностической практике как методы определения разных патогенов, главным образом вирусов. Предложенный нами новый метод изотермической амплификации основан на использовании амплификации, опосредованной никующим ферментом, в которой ДНК-дуплекс содержит повторы, позволяющие ему амплифицироваться неограниченное число раз без использования праймеров. Мы использовали новый метод изотермической амплификации ДНК для сайт-направленного мутагенеза участка искусственного интрона.

Annotation. Isothermal amplification of DNA is common clinical diagnostic methods of detection of various pathogens, mainly viruses. The new method of isothermal amplification based on nicking enzyme-assisted amplification and ends repeats of DNA  which allowing to amplification an unlimited number of times without the use of primers is proposed. We used the new method of isothermal DNA amplification for site-directed mutagenesis of sequence of artificial intron.
